چکیده

We consider technical approaches to the problem of making 3-D images of large (>10 μm) life-science samples with about 10 nm resolution . We find no existing methods and discuss the possibility of using soft x-ray diffractive techniques especially holography. We propose a new form of Fourier transform x-ray holography using a reference object consisting of an array of small pinholes formed by etching nuclear tracks. This scheme promises high resolution and a simple way to determine both the phase and amplitude of the diffracted wavefield.

Induction of cancer stem-like cells in A549 cells after exposure to carbon ions and X-rays

Background: Cancer stem-like cells (CSCs) play a crucial role in the initiation, progression, and recurrence of cancer. Evidence indicates that the high linear energy transfer (LET) carbon ion beam is more effective against CSCs than the conventional X-ray beam. Carbon ion radiotherapy is considered as a promising cancer strategy, however, information about whether, or not, new CSCs are induced...

Single-shot femtosecond x-ray holography using extended references.

In the context of x-ray lensless imaging, we present a recent approach for Fourier transform holography based on the use of extended references. Major advances shown here rely on a high signal efficiency and on the direct image reconstruction of the object performed by a simple linear derivative. Moreover, the extended holographic reference is easy to manufacture and can be applied to a variety...

Propagation based phase contrast x-ray imaging of Caenorhabditis elegans

Introduction: Imaging of multicellular organisms with nano-scale resolution is one of the challenges of modern life sciences. To solve biological problems, often the combined information of several microscopy techniques is necessary. While confocal fluorescence microscopy and optical super-resolution fluorescence can detect the distribution of markers in live cells, electron microscopy (EM) off...

Three-dimensional x-ray Fourier transform holography: the Bragg case.

A novel approach to determine the structure of nanoscale crystals in three dimensions is proposed by the use of coherent x-ray Fourier transform holography in Bragg geometry. The full internal description is directly obtained by a single Fourier transform of the 3D intensity hologram. Together with the morphology, Bragg geometry gives access to the 3D displacement field within the crystal. This...
